Advanced therapeutics: the application of Prussian blue @ polyacrylic acid / gold aggregate nanoparticles in CT guided chemotherapy and enhanced photothermal therapy

In recent years photothermal therapy (PTT) has aroused extensive academic interest. PTT uses photothermal conversion agent (PTA) to generate enough heat under near infrared (NIR) light to ablate tumor. Compared with the traditional methods of tumor treatment such as surgery radiotherapy PTT has the advantages of minimally invasive less side effects high time space control ability. However PTT alone lacks the ability of complete tumor ablation. Therefore in order to obtain better therapeutic effect many researchers continue to explore PTA with enhanced photothermal conversion efficiency coordinated treatment with different therapies.

Prussian blue nanoparticles (Pb NPs) have been approved by the US Food drug administration as diagnostic therapeutic drugs due to their non toxicity. At present Pb based core-shell nanoparticles have been used in many fields as a kind of traditional heterostructure composites but they also have some shortcomings. For example core-shell nanoparticles coated with Pb usually have good photothermal properties but due to their smooth compact surface there is usually no drug loading space; when Pb is used as the core of core-shell nanoparticles because Pb NPs are encapsulated in other materials the photothermal properties of nanoparticles are usually greatly reduced.

Wang Tingting School of chemistry environmental engineering Changchun University of technology synthesized Prussian blue @ polyacrylic acid / gold polymer nanoparticles by a simple mild method( PB@PAA/Au-A JNPs)。 By controlling the ratio of water to isopropanol in the reaction system PAA spheres were selectively coated on one side of Pb NPs gold aggregates were then grown on PAA templates resulting in the double-sided structure of nanoparticles. Compared with the common core-shell structure its double-sided structure has great advantages: two bare domains provide more exposed surfaces which ensure the maximum light absorption of nanoparticles under NIR light its photothermal conversion efficiency has been significantly improved on this basis showing significantly enhanced photothermal performance; the particle's negative charge also gives the particles a certain drug loading capacity. PB@PAA/Au-A Jnps integrates enhanced PTT chemotherapy CT imaging into a single nano platform which realizes the integration of enhanced PTT chemotherapy diagnostic imaging for cancer solves the problem that ordinary core-shell nanoparticles can not enhance photothermal absorption load drugs at the same time. The results of this work are as follows PB@PAA/Au-A Jnps have opened up new opportunities for tumor therapy will contribute to the development of a new generation of tumor therapeutic nanomaterials.
